Labuschagne went to school at Brisbane State High School before his family moved to Australia in 2004 when he was 10 years old as a result of his father finding employment in the mining sector.

He spoke Afrikaans growing up and only learned to speak English well after relocating to Australia.

Labuschagne, a right-handed batter, represented Queensland at the under-12, under-15, under-17, and under-19 age groups, serving as team captain during the 2012–13 National Championships. He represents the Easts-Redlands District Cricket Club in Brisbane Grade Cricket.

A prolific scorer for both teams, he played club cricket in England for Plymouth in the Devon Premier League in 2013 and Sandwich Town Cricket Club in the Kent Premier League in 2014.

In October 2015, Labuschagne made his List A debut for Queensland in the 2015 One-Day Cup. The following month, in his eighth Sheffield Shield game, he hit his first-ever first-class century.

Labuschagne committed to play for Glamorgan County Cricket Club in the 2019 English cricket season in April. In his maiden County Championship season, he scored 1,114 runs, including three hundreds in his first four first-class games, one of which was a then-career-best 182 against Sussex in a Glamorgan record second-wicket partnership of 291.

He was included in Australia’s Test team in September 2018 for their matchup with Pakistan. On October 7, 2018, he played in his maiden Test match for Australia against Pakistan, making a two-ball duck in the first innings and 13 runs in the second. On his debut, he also had two wickets.

As a result of a change in the International Cricket Council’s (ICC) rules, Labuschagne replaced Steve Smith on day five of the second Test on August 18, 2019, after Smith had sustained a concussion the day before. He was the first player to serve as a concussion substitute in a Test match.

Labuschagne scored 185, his first Test century, at the Gabba to keep his spot in the Australian lineup for Pakistan’s visit the following summer.

For their 2020 tour of India, Australia’s ODI team named Labuschagne in December 2019. On January 14, he played in his first one-day international; Australia won the game by 10 wickets, and Labuschagne did not bat or bowl.

Following the Test series, New Zealand left Australia and will play three more matches in 2020 on March 13, 15, and 20. Australia won the opening game by a score of 71 runs thanks to Labuschagne’s 56-run performance.

Labuschagne reached the milestone of 3,000 runs in December 2022 while the West Indies were touring Australia. He did it by becoming the joint second-fastest batter to do so. He needed 51 innings to reach the milestone, matching West Indies hitter Everton Weekes.

Labuschagne was a member of the ICC Men’s Test Team of the Year for the calendar year 2021 in the annual ICC Awards held in January 2022.
